Probing Biochemical Differences in Lipid Components of Human Cells by Means of ATR-FTIR Spectroscopy

Infrared spectroscopy has emerged as a promising technique for studying the composition of biological samples like lipids that play important roles in cellular functions and are involved various diseases. For this reason, target interest many biomedical studies. The objective present study is to utilize Fourier-Transform (FT-IR) examine main lipid components human cells (phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, phosphatidylinositol, C18 ceramide, sphingosine-1-phosphate, ceramide-1-phosphate, sphingomyelin, cholesterol, triolein). FT-IR analysis on previously mentioned was performed Attenuated Total Reflection (ATR) mode. obtained spectra clearly evidence contributions different functional groups examined samples. Detailed assignments spectral features were carried out agreement with literature. Similarities differences among types commercial evidenced discussed, particular attention phospholipid sphingolipid components. A quantitative phosphatidylinositol sphingomyelin using ratiometric approach reported. Moreover, reconstruction procedure complex useful chemometrics applications described. These representative examples potential use results can certainly contribute larger lipidomics.
